Guanine’s sensor electrochemically quantifies nucleic acid targets and associated 5mC and 5hmC without bisulfate conversion, PCR or sequencing using electroactive oligonucleotide tags and an inexpensive potentiostat.. Nucleic acid targets bind to magnetic microparticles preconjugated with capture probes and millions of electroactive guanine-quadruplex oligonucleotide tags. After magnetic purification, the magnetic particle-target complexes form sandwiches on a sensor electrode coated with recognition probes. Nanoparticles with anti-5mC antibodies and adenine-quadruplex tags and nanoparticles with anti-5hmC antibodies and thymine-quadruplex tags selectively bind to methylation sites on the nucleic acid targets. A voltammetry scan produces electrical current peaks proportional to the quantities of targets, 5mC and 5hmC. The process is rapid, simple and inexpensive and can multiplex using different electrodes.
